Een host-microcontroller kan communiceren met en de nRF24L01+ configureren via een 4-pins Serial Peripheral Interface (SPI). De configuratieregisters zijn toegankelijk via de SPI-verbinding. Configureerbare parameters omvatten frequentiekanaal (125 selecteerbare kanalen), uitgangsvermogen en gegevenssnelheid (gegevenssnelheden: 250 kbps, 1 Mbps en 2 Mbps).
De spanningsregelaar op de chip accepteert voedingsspanningen van 1,9 tot 3,6 V.
De module heeft 5V-tolerante ingangen, waardoor SPI-pinnen rechtstreeks op de Arduino kunnen worden aangesloten.
